HMMDA16S - HMP Single Cell MDA 16S rRNA Sanger Sequencing
Currently, there are over 1000 bacterial reference genomes at various stages of sequencing within HMP, however we are still far from covering the breadth of phylogenetic diversity of human associated microorganisms. As part of the technology development goals of the HMP, we are exploring recent advances in single cell-based techniques, such as multiple displacement amplification (MDA), in order to access hard-to-culture members of the microbiome.
Bacteria were obtained from HMP approved fecal, oral and skin samples. Microbial cell fractions were purified, and sorted single cells were lysed, DNA amplified by multiple displacement amplification (MDA) and 16S sequenced using Sanger sequencing.
Here we provide forward and reverse 16s rRNA sequences, used to taxonomically classify the MDA reactions.
